The reaction was genuine, but I am not sure what the adjective genuine is describing in your question.

They didn't know it was fake. So the emotion that they felt was real.

Because it's a reaction the audience wasn't necessarily wanting or expecting to have. It came from their heart. However phony Kaufman's acts might have been, the reaction from the audience was genuine.
 
I saw Man on the Moon, and liked it very much. Kaufman seems like an interesting guy, and I wish that I could have seen some of his work while he was still alive. Of course, I love Monty Python as well, so perhaps that kind of humor just appeals to me.
